Description
The Delta-Notch pathway controls developmental processes including cell fate determination, terminal differentiation, and proliferation. In mammals, four Notch receptors (NOTCH1–4) and five activating canonical ligands (JAGGED1, JAGGED2, DLL1, DLL3 and DLL4) have been described. Delta-like 1 homolog (DLK1) is a noncanonical Notch ligand, and acts as an inhibitor of Notch signaling. The inhibitory effect of DLK1 and DLK2 on Notch signaling may be mediated by competition of with canonical ligands for the receptor binding site. Widely expressed during embryonic development, expression becomes restricted in the adult to (neuro)endocrine tissues like the pituitary gland, adrenal glands, pancreas, monoaminergic neurons in the central nervous system, testes, prostate, and ovaries.
