Description
The mouse monoclonal antibody clone 5E10 reacts with human CD90, also known as Thy-1 (thymus cell antigen-1), a 25–37 kDa GPI-anchored glycoprotein and the smallest member of the immunoglobulin superfamily. CD90 contains a single V-type Ig domain and, lacking an intracellular domain, signals through lateral interactions with Src family kinases, G-inhibitory proteins, and RGD-binding integrins within lipid rafts. CD90 is expressed on thymocytes, neurons, fibroblasts, activated endothelial cells, and a small subset of CD3+CD4+ T cells in peripheral blood. In the hematopoietic compartment, CD90 marks a subset of CD34+ prothymocytes and hematopoietic stem cells in bone marrow, fetal liver, and cord blood, where the CD90+CD34+ population is enriched for long-term culture-initiating cells. CD90 is also a defining surface marker of mesenchymal stromal cells (MSCs), included in the minimal criteria established by the International Society for Cell Therapy. CD90 participates in cell-cell and cell-matrix adhesion, T cell signal transduction, neurite outgrowth, and regulation of MSC differentiation capacity. This antibody is reported for use in flow cytometric analysis, immunoprecipitation, western blotting, and immunohistology of frozen tissue sections.
