Description
The mouse monoclonal antibody clone 104D2 binds to human CD117 (also known as c-Kit), a 145 kDa type 1 transmembrane receptor that serves as the receptor for Stem Cell Factor (SCF). CD117 is expressed on hematopoietic stem cells in the bone marrow, including progenitor cells and mast cells, and is used as a flow marker to identify hematopoietic progenitor cells, mast cells, melanocytes, and gastrointestinal stromal tumor cells (GISTs). It constitutes about 1-4% of bone marrow cells, with 50-70% co-expressing CD34. In contrast, B-lymphoid committed progenitor cells (CD34+ CD19+) have low levels of CD117. Among CD34- bone marrow cells, only a few cells, primarily erythroid cells, express the CD117 receptor. CD117/c-Kit is a tyrosine kinase receptor. Upon binding of SCF, CD117/c-Kit undergoes dimerization and autophosphorylation, activating crucial signaling pathways for hematopoietic stem cell proliferation and differentiation. The antibody reportedly does not interfere with the binding of c-Kit ligand or the SR1 antibody.

RECOMMENDED USAGE
Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µL per 1 million cells in a staining volume of 100 µL. If whole blood is analyzed, then use 5 µL per 100 µL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.
Please briefly centrifuge the reagent vial before use.
