Description
The mouse monoclonal antibody clone 2D1 binds to human CD45. Human CD45, also known as Leukocyte Common Antigen (LCA), is one of the most abundant hematopoietic markers. It is a 180-220 kDa single-chain type I transmembrane protein expressed at high levels on all cells of hematopoietic origin, except erythrocytes, platelets, and non-hematopoietic cells. Specifically, CD45 is found on the surface of T and B lymphocytes, natural killer cells, monocytes, macrophages, dendritic cells, neutrophils, eosinophils, and basophils. CD45 functions as a protein tyrosine phosphatase with several isoforms, each generated and expressed in cell-specific patterns. With its broad cell distribution, CD45 plays a critical role in many leukocyte functions by regulating signal transduction and cell activation associated with the T cell receptor, B cell receptor, and IL-2 receptor. Other forms of CD45 with more restricted cellular expression include CD45R (B220), CD45RA, CD45RB, and CD45RO. It is reported that clones 2D1 and HI30 anti-human CD45 clones can reciprocally block the binding of each other to CD45.

RECOMMENDED USAGE
Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.
Please briefly centrifuge the reagent vial before use.
