Description
The mouse monoclonal antibody clone HP-3D9 reacts with human CD94 (also known as KLRD1 or KP43), a ~25-43 kDa type II transmembrane glycoprotein and member of the C-type lectin superfamily. CD94 is expressed on the cell surface as a disulfide-linked heterodimer with members of the NKG2 family, forming functionally distinct receptor complexes depending on the NKG2 partner. CD94/NKG2A and CD94/NKG2B heterodimers function as inhibitory receptors bearing cytoplasmic ITIMs, whereas CD94/NKG2C and CD94/NKG2E form activating receptor complexes that signal through the DAP12 ITAM-containing adapter protein. All CD94/NKG2 complexes recognize the non-classical MHC class Ib molecule HLA-E, which presents peptides derived from signal sequences of classical MHC class I molecules. CD94 expression is highest on NK cells, particularly activated NK cells, and is also found on NKT cells, γδ T cells, and a subset of CD8+CD56+ αβ T cells. Crossblocking studies indicate that the HP-3D9 clone recognizes the same epitope as the DX22 clone, and this antibody has been reported to inhibit the cytolytic activity of activated NK cells. This antibody is reported for use in flow cytometric analysis.
