Description
The 581 monoclonal antibody reacts with human CD34, a ~110–115 kDa monomeric type I transmembrane phosphoglycoprotein expressed on hematopoietic stem and progenitor cells (HSPCs), mesenchymal stem cells, embryonic fibroblasts, vascular endothelial cells, and some tumor cell lines. CD34 expression is transient during hematopoiesis and is gradually lost as progenitor cells mature. CD34 is widely used as a clinical marker for the identification, enumeration, and isolation of HSPCs for bone marrow transplantation. CD34 is thought to mediate attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells during early hematopoiesis, and to be involved in lymphocyte recruitment through binding to L- and E-selectin. The 581 clone recognizes a class III CD34 epitope that is resistant to neuraminidase, chymopapain, and glycoprotease. This antibody is reported for use in flow cytometric analysis and immunohistochemical applications.

RECOMMENDED USAGE
Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.
Please briefly centrifuge the reagent vial before use.
