The rat monoclonal antibody clone HEK/1/85a binds to human CD195, also known as CCR5. CCR5 is a 45 kDa G protein-coupled receptor (GPCR) featuring seven transmembrane domains. It belongs to the beta chemokine receptor family and is primarily expressed on subsets of T lymphocytes (particularly CD3+CD45RO+CD95+ cells) and monocytes. CCR5 binds several chemokines, notably MIP-1α, MIP-1β, RANTES, and MCP-2, triggering intracellular signaling pathways essential for cell differentiation, proliferation, chemotaxis, lymphocyte activation, and transendothelial migration during inflammatory responses. CCR5 is also recognized as a co-receptor necessary for the entry of macrophage-tropic HIV-1 (R5 HIV-1) into CD4+ immune cells. Post-translational modifications, such as sulfation, significantly influence HIV entry efficiency. Genetic studies highlight that individuals with partial or complete CCR5 gene deletions exhibit marked resistance to HIV infection. Additionally, CCR5 is implicated in various human diseases, including infections, inflammation, and cancers.

RECOMMENDED USAGE

Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.
