The mouse monoclonal antibody clone HB7 binds to human CD38, a 45 kDa type II single-chain transmembrane glycoprotein. CD38, also known as T10, is expressed at the highest levels on plasma cells, but expression can be found on activated T cells and terminally differentiated B cells. Moreover, the expression of CD38 during B cell development is bimodal, exhibiting high levels in immature cells, decreasing in intermediate cells, and increasing again in mature B cells. CD38 is also present in various tissues and hematopoietic cells, including T cells, NK cells, and monocytes. It can also serve as a marker for phenotyping leukemias and monitoring HIV-1 progression. The CD34+CD38- population of hematopoietic stem cells is considered to represent the most pluripotent cells, such as blast colony-forming cells. CD38 has also recently been identified in the nucleus, where it may function in the regulation of calcium levels in addition to its surface expression. CD38 classically has two functions: a receptor or an enzyme. As a receptor, CD38 binds to CD31. As an ectoenzyme, CD38 functions both as ADP-ribosyl cyclase and cyclic ADP-ribose hydrolase to catalyze the synthesis of cyclic ADP-ribose to regulate intracellular calcium level, thus playing an essential role in cell signaling and growth. Additionally, CD38 is a prognostic marker for some leukemias and other diseases.

RECOMMENDED USAGE

Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.

Please briefly centrifuge the reagent vial before use.
