The rabbit monoclonal antibody clone SP126 recognizes human inducible nitric oxide synthase (iNOS), also known as NOS2. iNOS is a ~130 kDa cytoplasmic enzyme belonging to the nitric oxide synthase family that catalyzes the production of nitric oxide (NO) from L-arginine. Unlike constitutively expressed nitric oxide synthases, iNOS expression is induced in response to inflammatory stimuli such as interferon-γ, tumor necrosis factor-α, and microbial products. iNOS is predominantly expressed in activated macrophages, dendritic cells, and other inflammatory cell types, where nitric oxide functions as an antimicrobial effector molecule and regulator of immune signaling. Detection of iNOS is widely used for characterization of inflammatory activation states, macrophage polarization, and immune responses in infection, cancer, and autoimmune disease. Clone SP126 is suitable for intracellular staining following fixation and permeabilization and is validated for flow cytometry and immunohistochemical applications.

RECOMMENDED USAGE
Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.
