The mouse monoclonal antibody clone WT31 binds to a conformational epitope formed by the human alpha/beta T cell receptor (TCR αβ) in complex with the CD3 epsilon chain. The TCR αβ heterodimer is composed of a 44 kDa alpha chain and a 37 kDa beta chain, whose specificity is determined by V alpha, J alpha, V beta, D beta, and J beta gene rearrangement. The TCR αβ heterodimer associates non-covalently with the CD3 complex, which is composed of at least five polypeptides including CD3 gamma, delta, epsilon, and zeta chains. Together, the TCR/CD3 complex recognizes antigen-derived peptides bound to MHC molecules on antigen-presenting cells such as B cells, dendritic cells, and macrophages, leading to T cell activation, intracellular signaling, and cytokine production. The TCR αβ heterodimer is expressed on the majority of peripheral blood T cells, while a minor population of CD3+ T cells expresses the alternative gamma/delta TCR heterodimer and is not recognized by WT31. The WT31 antibody has been reported to induce proliferation of resting T cells and to inhibit cytotoxic T cell activity. Some anti-CD3 antibodies partially inhibit WT31 staining, and clone SK7 is recommended for use in two-color experiments with WT31.

RECOMMENDED USAGE

Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.

Please briefly centrifuge the reagent vial before use.
