The mouse monoclonal antibody clone 5C3 binds to human CD40, a 45–50 kDa type I transmembrane glycoprotein and member of the tumor necrosis factor receptor (TNFR) superfamily, also known as TNFRSF5. CD40 is constitutively expressed on B lymphocytes at all stages of development except terminally differentiated plasma cells, and is also present on follicular dendritic cells, monocytes, macrophages, thymic epithelium, a subset of peripheral T cells, and some non-hematopoietic cells including endothelial cells and fibroblasts. CD40 functions as a receptor for CD154 (CD40L, gp39), which is predominantly expressed on activated CD4+ T helper cells. The CD40–CD154 interaction plays an essential role in T cell-dependent B cell activation, driving germinal center formation, immunoglobulin isotype class-switching, memory B cell development, and protection of B cells from apoptosis. Clone 5C3 has been validated for flow cytometric analysis of human peripheral blood mononuclear cells and has been reported for use in immunohistochemical staining of frozen tissue sections and in vitro B cell activation functional assays.

RECOMMENDED USAGE
Each lot of this antibody is quality control tested using flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 μL per 1 million cells in a staining volume of 100 μL. If whole blood is analyzed, then use 5 μL per 100 μL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application.
Please briefly centrifuge the reagent vial before use.
