It does not react with other classes of mouse immunoglobulin including IgD, IgG or IgA. IgM is expressed intracellularly, during early stages of B lymphopoiesis, and then on the surface of more mature B cells in the bone marrow and peripheral B cells. Fluorochrome conjugated II\/41 can be used as a detection secondary for mouse IgM. \u003c\/p\u003e\u003cp\u003eApplications Reported: The II\/41 antibody has been reported for use in flow cytometric analysis, immunohistochemical staining, and immunocytochemistry. \u003c\/p\u003e\u003cp\u003eApplications Tested: This II\/41 antibody has been tested by flow cytometric analysis of mouse bone marrow cells. This can be used at less than or equal to 1.0 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells\/test. This II\/41 antibody has been tested by immunocytochemistry of fixed cells using a mouse IgM primary antibody. This antibody can be used at less than or equal to 10 µg\/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. \u003c\/p\u003e\u003cp\u003eSuper Bright 780 is a tandem dye that can be excited with the violet laser line (405 nm) and emits at 780 nm. We recommend using a 780\/60 bandpass filter. Please make sure that your instrument is capable of detecting this fluorochrome. \u003c\/p\u003e\u003cp\u003eWhen using two or more Super Bright dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (\u003ca href=\"https:\/\/www.thermofisher.com\/order\/catalog\/product\/SB-4401-42\"\u003eProduct # SB-4401\u003c\/a\u003e) to minimize any non-specific polymer interactions. In such circumstances, we would recommend setting compensation with cells.
