The TCR αβ heterodimer is composed of a 44 kDa alpha chain and a 37 kDa beta chain, whose specificity is determined by V alpha, J alpha, V beta, D beta, and J beta gene rearrangement. The TCR αβ heterodimer associates non-covalently with the CD3 complex, which is composed of at least five polypeptides including CD3 gamma, delta, epsilon, and zeta chains. Together, the TCR\/CD3 complex recognizes antigen-derived peptides bound to MHC molecules on antigen-presenting cells such as B cells, dendritic cells, and macrophages, leading to T cell activation, intracellular signaling, and cytokine production. The TCR αβ heterodimer is expressed on the majority of peripheral blood T cells, while a minor population of CD3+ T cells expresses the alternative gamma\/delta TCR heterodimer and is not recognized by WT31. The WT31 antibody has been reported to induce proliferation of resting T cells and to inhibit cytotoxic T cell activity.
