CD34 expression is transient during hematopoiesis and is gradually lost as progenitor cells mature. CD34 is widely used as a clinical marker for the identification, enumeration, and isolation of HSPCs for bone marrow transplantation. CD34 is thought to mediate attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells during early hematopoiesis, and to be involved in lymphocyte recruitment through binding to L- and E-selectin. The 581 clone recognizes a class III CD34 epitope that is resistant to neuraminidase, chymopapain, and glycoprotease.
