This cell surface molecule is expressed on hematopoietic cells, endothelial, epithelial, as well as tumor cell lines such as Jurkat, U937, and MCF7. CD112 is a ligand for DNAM-1 (CD226), a leukocyte adhesion molecule expressed on natural killer (NK) cells. Interaction between CD112 and DNAM-1 promotes NK cytotoxic activity, leading to tumor cell lysis. In addition to its role in tumor suppression, CD112 plays a cooperative role in cell-cell adhesion, particularly in the formation of adherens junctions between neighboring epithelial cells and fibroblasts. CD112 can form homophilic and heterophilic interactions to regulate contact inhibition of cell movement and proliferation.
